Effect of novel curcumin nanoparticles on proliferation and apoptosis of Lewis lung cancer cells in vitro
Objective To determine the effect of novel curcumin nanoparticles (Cur-NPs) on the proliferation and apoptosis of Lewis lung cancer (LLC) cells in vitro. Methods LLC cells were cultured with different concentrations of Cur-NPs and free curcumin (Cur) for 24 h
and the cell proliferation was evaluated by MTT assay. After 20 mol/L of Cur-NPs and Cur treatment on LLC cells for 24 h
the cell apoptosis
cell cycle arrest
reactive oxygen accumulation
intracellular Ca
2+
accumulation and mitochondrial membrane potential were analyzed by flow cytometry.
Results Cur-NPs remarkably enhanced Cur-induced proliferation inhibition of LLC cells in vitro
and IC
50
decreased from 34.91 to 10.65 mol/L (P0.05). Cur-NPs significantly improved Cur-induced apoptosis (P0.05)
arrested cell cycle at S phase (P0.05)
increased reactive oxygen accumulation (P0.05) and intracellular Ca
2+
accumulation (P0.05)
and decreased mitochondrial membrane potential (P0.05). Conclusion Cur-NPs can enhance Cur-induced proliferation inhibition in LLC cells
which may be related to promotion of cell apoptosis.
