摘要

目的探讨PCDH10(protocadherin 10)基因对多发性骨髓瘤细胞系RPMI-8226增殖、迁移和侵袭能力的影响及其相关机制。方法采用脂质体将pcDNA3.1(+)PCDH10 及pcDNA3.1(+)质粒稳定转染入骨髓瘤RPMI-8226细胞，分别为PCDH10转染组、空质粒转染组，未转染组作为对照。采用RT-PCR及Western blot检测转染前后PCDH10基因和蛋白的表达，应用MTT法检测转染前后细胞的增殖能力；应用Transwell小室实验检测转染前后细胞的迁移及侵袭能力；应用Western blot检测增殖、迁移和侵袭相关蛋白表达水平。结果 PCDH10基因转染RPMI-8226细胞后，PCDH10在基因和蛋白水平均呈现高表达。MTT结果显示，PCDH10转染组增殖能力较对照组减弱；Transwell实验结果显示PCDH10能抑制瘤细胞的迁移、侵袭能力，PCDH10转染组迁移、侵袭的细胞数分别为(51.00±2.65)、( 51.00±8.00 )个，与对照组比较显著减少(P0.05)；Western blot检测结果显示PCDH10基因可下调AKT、p-AKT、cyclinD1和MMP-9的表达(P0.05)。结论 PCDH10基因可能通过下调PI3K/AKT通路活性而抑制多发性骨髓瘤细胞RPMI-8226的增殖、迁移和侵袭。

Abstract

Objective To investigate the effects of protocadherin 10 (PCDH10) gene on proliferation

migration and invasion of multiple myeloma (MM) cell line RPMI-8226 and its mechanism. Methods The RPMI-8226 cell line was transfected with pcDNA3.1(+)PCDH10 and pcDNA3.1(+) by lipofectamine and named as PCDH10 transfected group and empty vector group

respectively

with untreated cells as control. Real-time PCR and Western blot was used to investigate mRNA and protein expression of PCDH10. MTT assay was employed to determine cell proliferation. Transwell chamber and Matrigel assay were applied to observe the ability of migration and invasion. Western blotting was used to detect the expression of related proteins. Results The levels of mRNA and protein of PCDH10 were highly expressed in the PCDH10 transfected group. MTT assay showed that the proliferation of PCDH10-8226 cells was suppressed in the PCDH10 transfected group. The numbers of migration and invasion cells the PCDH10 transfected group were 51.00±2.65 and 51.00±8.00

respectively

significantly less than those from the control group (P0.05). Western blot results showed that PCDH10 could down-regulate the expressions of AKT

p-AKT

cyclinD1 and MMP-9 (P0.05). Conclusion PCDH10 gene inhibits proliferation

migration and invasion in MM cell line RPMI-8226

which may involves PI3K/ATK pathway.
