Objective        To construct a transposon (Tn917) library of Staphylococcus. aureus (S. aureus) Newman strain

and then identify the mutants with altered persistence.       Methods        Plasmid pID408 with transposon Tn917 was transformed into S. aureus Newman strain by electroporation

and high temperature and erythromycin were employed to induce transposition. Then a transposon library was constructed

and its quality was evaluated. After treating with 100MIC daptomycin for 24 and 36 h

2 specific mutant strains (T18 and T18s) were screened out

and their persistence were compared with that of wild type.       Results        The S. aureus Tn917 library was constructed

and 19 strains out of the library were randomly picked to identify their insertion sites. The results showed a good randomness. At the time point of 24 and 36 h

the wild type

T18 and T18s strains presented different persistence to some extent.       Conclusion        A S. aureus transposon library with good randomness is successfully constructed

which could be used for various purposes. And 2 specific mutant strains with altered persistence are screened out from it. The wild type

T18 and T18s strains show persistence difference in 24 and 36 hours’ culture.
