Effect of chronic intermittent hypoxia on NF-κB and TNF-α expression in cortex of rat after cerebral ischemia reperfusion injury
Objective To study the roles of chronic intermittent hypoxia (CIH) in the expression of inflammatory cytokines
nuclear factor-κB (NF-κB) p56 and tumor necrosis factor-α (TNF-α)
in the brain cortices of rats with cerebral ischemia reperfusion (CIR) injury. Methods In this experiment
36 Sprague-Dewley rats were randomly divided into 4 groups
a control group
a CIH group
a CIR group
and a CIH+CIR group. The rat model of CIH was made
and then the rats were exposed to CIR injury. The effusion of polymorphonuclear leucocytes in the brain tissue
was detected by HE staining. The expression of NF-κB p65 and TNF-α in the brain cortex was detected by immunohistochemistry assay. Results The expression of NF-κB p65 and TNF-α in the cortices of rats was significantly higher in the CIH group
the CIR group and the CIH+CIR group than in the control group (
P
0.05). Moreover
there was an interactive activity between CIH and CIR (
P
0.05). The expression of NF-κB p65 and TNF-α in the cortices of rats was significantly higher in the CIH+CIR group than in the CIH group and the CIR group (
P
0.05). The expression of NF-κB p65 was positively correlated with the expression of TNF-α (r=0.90
P
0.01). Conclusion CIH increases the expression of NF-κB p65 and TNF-α in the cortices of rats with CIR injury.
