摘要

目的对肺癌耐药相关基因CA916798的核心启动子区域进行初步鉴定。方法采用PCR方法对CA916798基因上游不同长度的序列片段0H～9H进行扩增，以pGL3-basic载体构建基因报告质粒，转染COS7细胞，荧光素酶检测序列启动活性，酶切鉴定、测序及数据库比对确定核心启动子的可能存在位置，并采用生物信息学方法对核心启动子区域结合位点进行预测。结果成功构建了启动子荧光素酶报告基因；活性实验表明；9H组与阴性对照组比较，相对荧光强度差异具有统计学意义(

0.05)；0H～8H各组与9H组比较，相对荧光强度差异均具有统计学意义(

0.05)；9H片段为处于CA916798基因片段上游179 bp处，长度为568 bp的序列区；功能预测显示该区存在多种细胞因子的结合位点。结论 9H序列区域具有较强的启动活性，为基因CA916798的核心启动子序列区域。

Abstract

Objective To identify the core promoter region of lung cancer drug resistance-related gene CA916798 preliminarily. Methods The upstream sequence 0H to 9H containing different fragments of CA916798 were amplified by PCR

and then the products were cloned into pGL3-basic plasmid. Then the obtained plasmids were transfected into COS7 cells. Promoter activities were evaluated by luciferase test. The possible position of core promoter was identified by endonuclease digestion

sequencing and BLAST. The binding sites of core promoter region were predicted by bioinformatics. Results Target fragments were successfully cloned.

The relative fluorescence intensity of group 9H was significantly higher than negative group (

0.05). As compared with group 0H to 8H

the relative fluorescence intensity were also markedly increased in group 9H (

0.05). 9H fragment was 568 bp

which located at 179 bp upstream of CA916798. The binding sites for several transcriptional factors were detected in the process of function prediction. Conclusion 9H fragment has a strong promoter activity

and the sequence is the major transcriptional locus of CA916798.
