Expression of programmed cell death gene 4 in triple-negative breast cancer and its relationship with prognosis
Objective Programmed cell death gene 4 (PDCD4) is a tumor suppressor gene
and plays an important role in regulation of cell apoptosis. The study was to investigate the expression of PDCD4 and its correlation with clinicopathological characteristics and prognosis of triple negative breast cancer. Methods A total of 176 patients with pathologically diagnosed triple-negative breast cancer and c
omplete follow-up data were subjected from Department of Breast Surgery in Guangdong Maternal and Children Hospital during Jan
2007 to Dec
2012
and all patients were women
with a median age of 44. PDCD4 expression in the 176 specimens and 10 specimens of para-tumor breast tissues was detected by S-P immunohistochemical assay. SPSS 13.0 statistical software was used to conduct Chi-square test
and Kaplan-meier and COX proportional hazards model was used to analyze the prognosis. Results The positive rate of PDCD4 in the tumor cells of triple negative breast cancer was significantly lower than that in the para-tumor breast tissues (44.8% vs 90.0%
P
=0.005). The loss of PDCD4 expression was closely correlated to tumor size (r=-0.270
P
0.001)
lymph node metastasis (r=-0.240
P
=0.001) and histological grade (r=-0.206
P
=0.006). The median follow-up period was 42 months. The disease free survival and overall survival rate was significantly higher in PDCD4-positive group than in PDCD4-negative group (
P
0.05). Cox model revealed that PDCD4 expression
tumor size
and stage were independent prognostic factors of triple-negative breast cancer (
P
0.05). Conclusion PDCD4 expression is down-regulated in triple-negative breast cancer
and the loss of PDCD4 expression is correlated to poor prognosis. PDCD4 is an independent prognostic factor of triple-negative breast cancer.
