Effect of ovarian cancer cells on MMP-2 and MMP-9 expression and tube formation of endothelial progenitor cells in co-culture system
Objective To observe the effect of vascular endothelial growth factor on the capacity of tube formation and expression of MMP-2 and MMP-
9 in endothelial progenitor cells (EPCs) after co-cultured with ovarian cancer cell line SKOV3. Methods Density gradient centrifugation was used to isolate EPCs. The capacity of tube formation was determined in EPCs that was cultured in Martigel (EPCs group) and co-cultured with SKOV3 cells (co-culture group)
respectively
and the tube formation capacity of EPCs was also determined after adding different concentrations of anti-VEGF antibody in the co-culture system. RT-qPCR and Western boltting were applied to determine the expression of MMP-2/MMP-9 in EPCs. Results The average number of tube formation in the EPCs group (18.00±1.00) was significantly lower than that in the co-culture group (36.33±1.52) (
P
0.05). The average numbers of tube formation in the co-culture group added with 25
50 and 100 μg of anti-VEGF antibody were 32.00±1.00
26.33±0.57 and 20.33±1.15
respectively
which were all less than that in the co-culture group without anti-VEGF antibody (
P
0.05). The results of RT-qPCR and Western blotting showed that the co-culture group showed an increase in the mRNA and protein expression of MMP-2 and MMP-9 as compared with the EPCs group by 4.36 fold (
P
0.05)
3.34 fold (
P
0.05) and 2.99 fold (
P
0.05)
and 6.63 fold (
P
0.05)
respectively. Adding with 100 and 50 μg anti-VEGF antibody into the co-culture group resulted in an inhibition in the mRNA and protein expression of MMP-2 by 2.08 fold (
P
0.05) and 2.11 fold
P
0.05)
and 2.12 fold (
P
0.05) and 1.71 fold (
P
0.05)
respectively. Adding 100 μg anti-VEGF antibody into the co-culture group resulted in an inhibition in the mRNA and proteins expression of MMP-9 by 2.13 fold (
P
0.05) and 2.94 fold (
P
0.05)
respectively. Conclusion SKOV3 cells may regulate the expression of MMP-2 and MMP-9 in EPCs by secreting VEGF
thereby promoting the tube formation capacity of EPCs.
