Effect of budesonid inhalation on expression of TGF-β and CTGF in serum and BALF of bleomycin-induced pulmonary fibrosis rats
Objective To investigate the effect of budesonid inhalation on the expression of transforming growth factor (TGF)-β and connective tissue growth factor (CTGF) in serum and bronchoalveolar lavage fluid (BALF) of bleomycin-induced pulmonary fibrosis rats. Methods Forty-five SD rats were randomly divided into three groups (15 rats in each group) including a model group
a control group and an intervention group. Five rats in each group were randomly killed on the 7th
14th and 28th day after the intervention. HE staining and Masson staining were used to determine the degree of alveolitis and pulmonary fibrosis. The expression levels of TGF-β and CTGF in serum and BALF were determined by ELISA. Results (1) HE staining and Masson staining of lung tissues proved that the classical model of pulmonary fibrosis was successfully constructed. (2) The degree of alveolitis and pulmonary fibrosis in the model group and intervention group was significantly more serious than that in the control group (P0.05)
and the pathological changes in the intervention group were significantly improved as compared with the model group (P0.05). (3) The ELISA results showed the expression levels of CTGF and TGF-β were different in BALF and serum in each group
and the expression levels of CTGF and TGF-β in BALF were significantly higher than those in serum. (4) The expression levels of CTGF and TGF-β in the model group and intervention group were significantly higher than those in the control group (P0.05)
and they were significantly lower in the intervention group than in the model group (P0.05). Conclusion Budesonid can improve bleomycin-induced pulmonary fibrosis by reducing the expression levels of TGF-β and CTGF in BALF and serum of rats.
