Role of LOX-1 on tissue factor expression in human umbilical vein endothelial cells induced by C-reactive protein
Objective To investigate the effect of lectin-type oxidized LDL receptor 1(LOX-1)on the expression of tissue factor (TF) induced by C-reactive protein (CRP) in human umbilical vein endothelial cells(HUVECs). Methods HUVECs were transfected the recombination plasmid pTracer CMV2-CRP. The cells were divided into a normal control group
GFP group and CRP-GFP group. RT-PCR was used to detect the mRNA of TF and LOX-1
and Western blotting was used to detect the protein expression of LOX-1
TF
ERK1/2 and JNK respectively. After HUVECs transfected with CRP-GFP were treated with ERK1/2 inhibitor (U0126) or JNK inhibitor (SP600125) respectively
the expression of TF gene was detected by RT-PCR and Western blotting. Results CRP transfection into human HUVECs resulted in significantly increase in the expression of LOX-1 at mRNA and protein levels (0.70±0.03 vs 0.24±0.01
0.661 6±0.020 0 vs 0.255 8±0.040 0
P0.05) and the expression of TF (0.442±0.040 vs 0.146 0±0.030 0
0.727 4±0.020 0 vs 0.217 4±0.020 0
P0.05). CRP activated ERK1/2
but not JNK MAP kinase (MAPK)
and the effect of CRP on TF expression was blocked by pharmacological inhibitor of ERK1/2 (U0126)
but not JNK (SP600125) MAPK. Neutralizing antibody to LOX-1 significantly attenuated the mRNA and protein expression of TF induced by CRP (0.264 0±0.030 0 vs 0.442 0±0.040 0
0.324 4±0.040 0 vs 0.727 4±0.020 0
P0.05). Conclusion CRP enhances LOX-1 expression in HUVECs and thus promotes TF expression through LOX-1 receptor
by a underlying mechanism of ERK1/2 MAPK but not JNK MAPK pathway.
