Exogenous hydrogen sulfide improves epilepsy-induced cognitive dysfunction in rats
Objective To investigate the neuroprotective effect of exogenous hydrogen sulfide (H
2
S) on cognitive deficits induced by epilepsy in rats. Methods The epilepsy rat model was induced by intraperitoneally administration of LI-PILO. Male SD rats were divided into 3 groups
normal control group (NC group)
epilepsy group (EP group)
EP+NaHS group (NaHS group
intraperitoneally injection of 14 μmol/kg NaHS
in 25 min before PILO administration). The ability of spacial learning and memory of epilepsy rats was tested by Morris water maze. The expression of cystathionine-β-synthase (CBS) and cAMP response element binding protein (CREB) in the hippocampus was observed by real-time PCR in 2 and 24 h
and 7 and 30 d after model establishment. Results Morris water maze test showed that the spatial learning and memory ability of EP rats were decreased when compared with NC group. In EP rats
the escape latency to find the platform was significantly prolonged or extended compared with NC group (P0.01). The times of passing platform
mean exploring time and percentage of swimming distance in original platform quadrant were decreased significantly compared with NC group (P0.01). NaHS improved the ability of learning and memory
with all indices of spatial learning and memory had significant differences when compared with EP group (P0.05
0.01). Compared with NC group
the expression of CBS mRNA in the hippocampus of EP rats were down-regulated (P0.05) in 7 and 30 d. While
the expression of CBS mRNA in NaHS group was higher than EP (P0.05). Compared with NC group
the expression of CREB mRNA of EP rats were down-regulated in chronic period (7 and 30 d
P0.05). The expression of CREB mRNA in NaHS group was higher than EP group at 24 h
and 7 and 30 d (P0.05)
but had no statistical significance at 7 d and 30 d when compared with NC group (P0.05). Conclusion Exogenous H
2
S exerts neuroprotection effects and improves cognitive deficits in epileptic rats
which may be through upregulating the expression of CBS and CREB mRNA.
