Effect of medium molecular weight hydroxyethyl starch on neuropathic pain in rats after spinal cord injury
Objective To investigate the effect of medium molecular weight hydroxyethyl starch (HES 130/0.4
Voluven) on below-level neuropathic pain after spinal cord injury (SCI). Methods Using random number table
a total of 72 male adult SD rats were randomly and equally divided into four groups. Rats in three groups underwent moderate spinal cord contusion (T
10-11
) and were given normal saline
low molecular weight HES (HES 40) and me
dium molecular weight HES (HES 130/0.4
Voluven) through caudal vein
respectively; the other group of rats only underwent laminectomy and were set as control. In behavioral test
paw withdraw pressure threshold (PWPT) were measured to evaluate the central pain. Evans blue (EB) uptake in spinal cord tissues was measured to evaluate the disruption of blood spinal cord barrier. Levels of GFAP
Iba1 and ED1 were measured by Western blotting for the detection of astrocyte and macrophage/microglia activation. ELISA was applied to determine spinal expressions of IL-1
IL-6 and TNF-α. Results EB uptake in spinal cord tissues demonstrated that HES 130/0.4 effectively retrieved the compromised BSCB after 3 d of SCI as compared with normal saline and low molecular weight HES (Saline: 2.58±0.30; HES 40: 2.38±0.48; HES 130/0.4: 1.38±0.35; P0.05). Ten days after SCI
as compared with normal saline and low molecular weight HES
immunohistochemistry results showed that HES 130/0.4 significantly lessened macrophage/microglia activation, and ELISA showed HES 130/0.4 significantly reduced expressions of IL-1
IL-6 and TNF-α (P0.05). Behavioral test after ten-day SCI suggested that PWPT was dramatically reduced in all groups of rats
but HES 130/0.4 treatment could significantly raise the PWPT as compared with normal saline and low molecular weight HES (P0.05). Conclusion HES 130/0.4 could attenuated below-level neuropathic pain after spinal cord injury
and the mechanism maybe related to remedy of BSCB disruption and inhibition of macrophage/microglia activation.
