Acute toxicity and genetic toxicity of hogwash fat
Objective To determine edible safety of hogwash fat by toxicological methods. Methods Acute toxicity test
genetic toxicity test (mouse bone marrow micronucleus test
mouse sperm abnormality test) and 30-day feeding trial were carried out. Acute toxicity test was performed on 20 SD rats by being fed a one-time implementation (2.0 ml/100 g). A total of 60 Kunming mice were randomly divided into 6 groups
intragastric injection with 2.5
5.0 and 10.0 g/kg hogwash fat
norma
l saline
oil and cyclophosphamide (0.04 g/kg) respectively. These mice were killed in 30 h after injection
and their femur bone marrow fluid were obtained for mice bone marrow micronucleus test. Another 30 Kunming male mice were randomly divided into 6 groups as above mentioned and received intragastrical injection for 5 d. The mice were killed in 35 d later
and their bilateral epididymis was resected to observe sperm abnormalities. A total of 100 SD rats were randomly divided into 5 groups fed as mentioned above except cyclophosphamide group for 30 d
their body weight and food utilization
their hemogram and biochemical and pathological changes were observed. Results The hogwash fat used in acute toxicity had an oral LD
50
greater than 10.0 g/kg. Mouse bone marrow micronucleus test was positive
indicating that the hogwash fat had chromosomal damage on mouse bone marrow cells with mutagenicity. Mouse sperm abnormality test was negative. 30-day feeding test indicated that low dose hogwash fat led to slow weight gain and reduced food availability
high doses lead to reduced serum total protein and globulin content
and middle and high dose also led to an increase in the indexes for liver functions and liver pathological changes. Conclusion The hogwash fat used in the experiments is negative to acute toxicity
but positive to genetic toxicity tests and the 30-day feeding test
indicating that hogwash fat used in this experiment has toxic effects in experimental animals.
