Estrogens enhances the number of dopamine neurons of adult male mice though upregulating neurotrophic factors
) on dopamine neurons in the substantia nigra (SN) of adult male mice and the underlying molecular mechanism. Methods Ten C57/BL6 mice were randomized to 2 groups
control group (with subcutaneous injection of 0.1 ml sesame oil for 10 d) and E
2
group (with subcutaneous injection of 2 μg/d 17-β estradiol solved in 0.1 ml sesame oil for 10 d). Immunohistochemistry was used to detect the positive neurons of tyrosine hydroxylase (TH) and calbindin. The expression levels of dopamine transporters
(DAT)
insulin-like growth factor 1 receptor (IGF-1Rm) and brain-derived neurotrophic factor (BDNF) were detected by RT-PCR. Results E
2
increased the number of dopamine neurons [(119.7±2.9) vs (93.5±5.1)
]
(P=0.004) and the mRNA levels of DAT(P0.05)
but had no effect on the number of calbindin-positive neurons [(88.0±2.0) vs (86.2±1.5)
]
(P=0.507). E
2
increased the mRNA levels of IGF-1R and BDNF in the midbrain significantly(P0.05). Conclusion E
2
can increase the number of dopamine neurons through upregulating the expression of IGF-1R and BDNF.
