Relationship of telomerase activity with proliferation, migration and adhering of EPCs in peripheral blood in vitro
Objective To investigate the relationship of the activity of telomerase with the proliferation
migration and adhering of cultured endothelial progenitor cells (EPCs). Methods Mononuclear cells (MNCs) were harvested from human peripheral blood by density gradient centrifugation
then inoculated in culture plates coated with gelatin. After cultured for 7 d
the immunohistochemical method was performed to identify EPCs. The proliferation
migration and adhering of EPCs were measured by MTT assay
Transwell insert and the measured experiment of adhering ability. The activity of telomerase was detected by TRAP-PCR argentation. Results EPCs were well isolated from human peripheral blood. The proliferation
migration and adhering of EPCs and the activity of telomerase were reduced in patients with coronary heart disease (CHD)
compared with the control group. Conclusion The weakening of EPCs biological function might be correlated with the attenuation of telomerase activity.
