Expressions of muscle specific microRNAs in denervated skeletal muscle in mice
Objective To explore the expression changes of muscle special microRNAs (miR-1
miR-133 and miR-206) in the atrophying skeletal muscle induced by the denervation in order to establish a basis for further study on their roles in the denervated atrophying muscle. Methods The mouse model of sciatic nerve resection was prepared and evaluated by comparing the wet weight percentage of muscle before and after denervation and the cross section area (CSA)of the muscle fibers. Northern blotting was used to examine the levels of muscle special microRNAs expressions in the atrophying mouse gastrocnemius muscle at different time points after denervation. Results The mouse model of sciatic nerve resection was successfully established. The expressions of miR-206 was obviously up-regulated with the time of denervation
however
those of miR-1 and miR-133 was decreased firstly and then gradually recovered with the time elapse. Conclusion The changes of the expressions of the skeletal muscle special microRNAs is very striking
so we presume that they may play certain roles in the atrophying skeletal muscle resulted from denervation.
