Role of MLC20 phosphorylation in PKCα and PKCε regulating calcium sensitivity in cultured hypoxic VSMCs and in hemorrhagic shock rats
jective To observe the role of 20 kDa myosin light chain (MLC
20
) phosphorylation in the regulation of protein kinase Cα (PKCα) and PKCε on calcium sensitivity during hemorrhagic shock (HS) in rats for the mechanism of calcium desensitization. Methods Vascular smooth muscle cells (VSMCs) after hypoxia and superior mesenteric artery (SMA) from HS rats were adopted to observe the effects of PKCα and PKCε agonists on the activity of myosin light chain phosphatase (MLCP) and the phosphorylation of MLC
20
and observe the influence of inhibitors of PKC-potentiated inhibitory protein for protein phosphatase 1 of 17 kDa (CPI-17)
integrin-linked kinase (ILK) and zipper-interacting protein kinase (ZIPK) on the action of PKCα and PKCε agonists in these events by Western blotting and substrate phosphorylation. Results (1) MLCP activity was increased to 150.1% of normal control group in VSMC after 2 h hypoxia (P0.01)
and was decreased to 103.0% (P0.01) or 96.3%(P0.01) by PKCα agonist or PKCε agonist. Comparing with PKCα agonist group
MLCP activity was increased to 136.7% (P0.01)
127.6% (P0.01) and 116.7% (P0.01) after the treatment of the inhibitors of CPI-17
ILK and ZIPK respectively. Comparing with PKCε agonist group
MLCP activity was increased to 134.0%(P0.01)
128.5% (P0.01) and 126.0% (P0.01) respectively by the inhibitors of CPI-17
ILK and ZIPK. (2) MLC
20
phosphorylation was decreased from 32.4% to 10.4% (P0.01) in SMA after 2 h shock
and was increased to 26.4% (P0.01) and 25.6% (P0.01) by PKCα and PKCε agonists. Comparing with PKCα agonist group
MLC
20
phosphorylation was decreased to 12.7% (P0.01)
10.0% (P0.01) and 15.7% (P0.01) respectively by the inhibitors of CPI-17
ILK and ZIPK. Comparing with PKCε agonist group
MLC
20
phosphorylation was decreased to 10.1% (P0.01)
12.5% (P0.01) and 10.3% (P0.01) respectively by the inhibitors of CPI-17
ILK and ZIPK. Conclusion PKCα and PKCε may regulate the MLCP activity and MLC
20
phosphorylation via CPI-17
ILK and ZIPK
to modulate calcium sensitization of vascular smooth muscle following HS.
