Angiotensin Ⅱ induces adult adipose-derived mesenchymal stem cells to differentiate into cardiocyte-like cells
Objective To study the effects of angiotensin Ⅱ (AngⅡ)
as inducing agent in vitro
in inducing adult adipose-derived mesenchymal stem cells (ADMSCs) to the direction of myocardial cell. Methods ADMSCs were isolated and cultured from adult adipose tissues. Different concentrations of AngⅡ (0.05
0.10 and 0.20 μmol/L
respectively) were used to induce ADMSCs. Untreated ADMSCs were used as normal control. The cell morphological feature was observed under inverted phase-contrast microscope and transmission electron microscope. Immunocytochemical staining was used to detect the cardiac troponin-I (cTn-I) and calculate the conversion rate. Results After three weeks of induction
cTn-I-positive cells could be detected in 0.20 μmol/L AngⅡ group. Four weeks later
cTn-I-positive cells were detected in 0.10 and 0.20 μmol/L AngⅡ groups
furthermore
the number of cTn-I-positive cells in 0.20 μmol/L AngⅡ group was significantly higher than that of other groups. Conclusion Ang Ⅱ can induce in vitro ADMSCs to differentiate into cardiocyte-like cells
and the best induction concentration is 0.20 μmol/L.
