Objective To construct the replicate-deficient recombinant adenovirus containing Otos for further evaluation of its roles in cochlear.  Methods pAdTrack-Otos was cloned by inserting the fragment of Otos into the shuttle plasmid vector pAdTrack-CMV. For obtaining recombinant adenovirus plasmid (Ad-Otos)

the pAdTrack-Otos and the backbone plasmid pAdEasy-1 were homologously recombined in bacteria BJ5183

then pAdEasy-GFP-Otos was transfected into 293 cells for getting packaged infectious replicate-deficient adenoviruses.  Results By DNA sequencing

the fragment inserted within pAdTrack-Otos was verified as Otos. Ad-Otos efficiently infected 293 cells.  Conclusion Replicate-deficient recombinant adenovirus carrying Otos was successfully constructed

laying the experimental foundation for further studying the roles of Otos in inner ear.
