Evaluation and quantification of total antioxidative capability of fruit by microassay
Objective 〓To investigate the different kinetics and capability of natural antioxidants scavenging free radical 2
2-diphenyl-1-picrylhydrazyl (DPPH·). Methods Spectrum scanning was used to analyze the characteristic absorbance of DPPH. Microplate quantification of DPPH· was used to quantify total antioxidative capability (TAC) of vitamin C
chlorogenic acid
Chaenomeles
citrus
mango and cherry. TAC represents the amount (μg) of DPPH· scavenged by each gram or each milliliter of the samples. Results The highest absorbance of DPPH· was shown on microplate reader at wavelength of 490 nm. Vitamin C and chlorogenic acid verified good correlation between antioxidants concentration and amount of scavenging DPPH· by microplate quantification assay. The TAC of the fruit juices was as follows: Chaenomeles 87.23 (P0.05)
mango 62.556 (P0.05)
citrus 58.778 (P0.05) and cherry 49.072 (P0.05). Conclusion Microplate quantification assay reveals the characteristics of nature scavengers to complex compounds. It has the advantages of less reagents used
high throughput
easy accessibility to kinetic curve and minimized experimental error.
