摘要

目的探讨中药姜黄素对膀胱癌细胞(T24)增殖及凋亡的影响及可能机制。方法以培养的T24细胞为研究对象，姜黄素组加入终浓度为1、10、100 μmol/L的姜黄素，对照组不加姜黄素，12、24、48 h后观察 T24 细胞的形态变化，分别用MTT法、TUNEL染色、RT-PCR、免疫印迹及图像分析技术检测各组细胞生长、凋亡发生、AKT1蛋白及转录水平、caspase 3蛋白含量的变化。结果姜黄素组T24细胞生长抑制率显著上升，细胞凋亡发生显著升高，AKT1蛋白活性水平降低，转录水平无显著变化，caspase 3蛋白含量显著升高。结论姜黄素能抑制T24细胞的生长并促进其凋亡，其发生与其抑制T24细胞PKB/AKT信号通路有关。
