摘要

目的探讨过氧化物酶体增殖体激活受体-γ

（PPAR-γ

）基因外显子2的Pro12Ala多态性与肥胖的关系。方法运用多聚酶链式反应-限制性片段长度基因多态性（PCR-RFLP法）分析方法，对116例超重或肥胖患者和89例正常对照者PPAR-γ

基因Pro12Ala多态性位点进行基因分型。结果超重或肥胖组Ala等位基因频率(11.64%)显著高于正常对照组(5.06%)，在所检测人群中，PA/AA基因型者具有更高的体质量指数和血浆甘油三酯水平(P0.05)。结论 PPARγ

的Pro12Ala变异与肥胖的发生有关，12Ala更易发生肥胖。

Abstract

Objective To investigate the polymorphism of peroxisome proliferator-activated receptors-γ

(PPAR-γ

) and its association with obesity in Chinese population. Methods According to BMI

89 subjects who are in normal body weight range and 116 overweighted and obese subjects were included in this study. Their Pro12Ala mutation in the PPAR-γ

gene was detected by PCR restriction fragment length polymorphism. Results Allele frequency of Ala mutation of PPAR-γ

in overweighted and obese subjects (qA=11.64%) was significantly higher than that of normal body weight group (qA=5.06%

P0.05). Both in overweighted and obese subjects and normal body

weight subjects

these with genotype of PA (AA) had higher body mass index and plasma TG than those with the genotype of PP (P0.05). Conclusion Pro12Ala mutation in PPAR-γ

is associated with obesity. The people with the genotype of 12Ala are likely to be obese.
