摘要

目的探讨牵张负荷条件对膀胱Cajal间质细胞(interstitial cells of Cajal

ICCs)数量及c-kit蛋白表达变化的影响，阐明牵张负荷对膀胱ICCs细胞兴奋性的调控作用。方法构建大鼠不稳定逼尿肌模型，以光镜计数及免疫组化法检测牵张负荷下逼尿肌稳定组和逼尿肌不稳定组与正常对照组膀胱ICCs细胞数量及酪氨酸激酶受体(c-kit)在蛋白水平表达的差异。结果逼尿肌稳定组和逼尿肌不稳定组ICCs细胞数量与c-kit表达量较正常对照组均有增高(P001)，逼尿肌不稳定组较稳定组升高显著(P0.01)。结论牵张负荷对ICCs细胞有调控作用； ICCs细胞数量及c-kit表达增多可能是其机制之一。

Abstract

Objective To investigate the effects of stretch load on the quantity of rat bladder interstitial cells of Cajal (ICCs) and ckit expression in the cells. Methods Rat models of detrusor instability with partial outflow obstruction were established. The quantity of ICCs and c-kit expression in ICCs from stable

unstable and normal detrusor were studied with light microscopy and Western blotting. Results The quantity of ICCs and expression of c-kit in the stable and unstable detrusors were higher than that in normal control group (P0.01)

and those of the unstable detrusor were significant higher than those of the stable one (P0.01). Conclusion Stretch load can regulate the function of ICCs

probably through increment of the quantity of ICCs and up-regulation of c-kit expression in the cells.
