Abstract
Objective To study hematopoiesis reconstruction in mice of hematopoietic aplasia by transplanting hematopoietic stem/progenitor cells mobilized by Angelica polysaccharides (APS). Methods The female BALB/c mice irradiated with 8.5 Gy
60
Co γ ray were transplanted with peripheral blood mononuclear cells (PBMCs) from male BALB/c mice mobilized by APS. After transplantation
the changes of WBC
HGB and platelet in peripheral blood were observed. Thirty-day survival rate was calculated. PCR was used to verify the cell source of hematopoiesis reconstruction. Res
ults The number of WBC
platelet and HGB
and 30-day survival rate of APS group were markedly higher than that of negative control group and normal saline group (P0.05). PCR results showed Y chromosome sequence existed in the survived mice of APS group
indicating that the repopulated hematopoietic cells came from the male donor. Conclusion The hematopoietic stem/progenitor cells mobilized by APS can reconstruct long-term hematopoiesis in mice of hematopoietic aplasia.
