Isolation and enrichment of fetal nucleated red blood cells by using N-acetylgalactosamine-specific lectin
Objective To isolate and enrich fetal nucleated red blood cells (FNRBCs). Methods Fetal cells were collected by gradient centrifugation from umbilical cord blood collected during delivery
then mixed with soybean agglutinin (SBA) of a serial concentration (10
20
30
40
50 μg/ml) to isolate and enrich FNRBCs by using N-acetylgalactosamine (GalNAc)-coated chamber slides. Results The best working concentration of SBA was 20 μg/ml
which resulted in most effectively isolating and enriching FNRBCs in umbilical cord blood
when the coating concentration of GalNAc was 100 μg/ml. The ratio of white blood cells to nucleated cells were the lowest value (27.7%)
and that of FNRBCs to nucleated cells were the highest value (3.7%). Conclusion GalNAc-specific agglutinin could be used to effectively isolate and enrich FNRBCs.
