Experimental study on biodistribution and PET imaging of 3’-deoxy-3’-18F-fluorothymidine in murine models of lung carcinoma and inflammation
Objective To investigate the biodistribution and positron emission tomography (PET) imaging of 3′-deoxy-3′-
18
F-fluorothymidine (
18
F-FLT) in murine models of lung carcinoma and inflammation
so as to evaluate the use of
18
F-FLT as a new PET tracer for diagnosis of
malignant tumor. Methods Sixteen T739 mice bearing the lung adenocarcinoma were randomly divided into two groups according to the different tracers. The biodistribution of mice for
18
F-FLT and
18
F-fluorodeoxyglucose (
18
F-FDG) was measured with well-gamma detector at 60 min after injection from tail veins. PET imaging was performed using two tracers. The whole process mentioned above was repeated in twelve T739 mice with inflammation. Results In the biodistribution study of
18
F-FLT with lung carcinoma models
considerable radioactive uptake by tumor was observed. The ratios of tumor/blood
tumor/muscle and tumor/lung were all above 2.0. The tumor PET images with
18
F-FLT and
18
F-FDG were clear. In the study with inflammation models
inflammation-to-muscle radioactivity ratio of
18
F-FDG (2.25±0.39) were higher than that of
18
F-FLT (1.26±0.19) (P0.01).
18
F-FDG accumulated in inflammatory tissues
whereas
18
F-FLT was not taken up by these tissues. Conclusion The uptake of
18
F-FLT in pulmonary malignant tissues is higher than that in normal tissues. Inflammatory tissues are invisible in
18
F-FLT PET images.
18
F-FLT is of good specificity to differentiate the two models.
