摘要

目的观察巨噬细胞集落刺激因子（macrophage colony stimulating factor

M-CSF）对肺腺癌细胞株A549的作用。方法观察肺腺癌A549细胞形态的变化。M-CSF对体外培养肺腺癌A549细胞的生长抑制作用采用MTT 检测法

用流式细胞仪检测细胞DNA含量

分析M-CSF对细胞周期的影响。采用RT-PCR检测M-CSF受体mRNA表达的变化。结果 M-CSF能显著抑制肺腺癌A549细胞的生长，并且呈浓度、时间依赖关系，在M-CSF浓度为10 ng/ml时有最大抑制效应。流式细胞仪检测分析显示M-CSF作用后G

期细胞增加。RT-PCR显示经M-CSF作用后M-CSF受体的mRNA表达水平有所下降。结论 M-CSF对肺癌细胞株A549在体外有抑制作用。
