摘要

目的探讨CO

气腹对人宫颈癌CaSki细胞抗凋亡能力的影响。方法建立腹腔镜CO

气腹体外模型，以不同压力（0、7、14 mmHg）的CO

气腹对人宫颈癌CaSki细胞处理不同时间（1、2、3、4 h），设未处理为对照组。采用RT-PCR、Western blot分别检测CO

气腹处理后各组细胞Survivin mRNA 和Bcl-2蛋白的表达情况。结果当压力为7、14 mmHg时，Survivin和Bcl-2的表达均随着气腹处理时间的延长呈上升趋势（P0.05）。当处理相同时间时，7 mmHg组Survivin和Bcl-2的表达量均明显高于其他压力组和对照组（P0.05）。结论在一定压力和作用时间范围，CO

气腹能提高宫颈癌细胞的抗凋亡能力，这可能是CO

气腹促肿瘤生长的机制之一。

Abstract

Objective To study the effect of CO

pneumoperitoneum on the anti-apoptotic capability of human cervical carcinoma cell line (CaSki). Methods The imitated CO

pneumoperitoneum was established in vitro. After CaSki cells were exposed to CO

pneumoperitoneum at 0

7 or 14 mmHg for 1

4 h

the expressions of Survivin mRNA and Bcl-2 protein were examined by RT-PCR and Western blot respectively. The untreated CaSki cells served as control. Results Under 7 or 14 mmHg

the expressions of Survivn and Bcl-2 had an increasing tendency w

ith the exposure time prolongation. Within the same exposure time

the expressions of Survivin and Bcl-2 under 7 mmHg were significantly higher than those under 0 or 14 mmHg or in control group. Conclusion Under certain pressure and within certain exposure time

pneumoperitoneum could improve anti-apoptotic capability of human cervical carcinoma cells

which may be one reason for the promoting effect of carbon dioxide pneumoperitoneum on malignant cells growth.
