Effect of nitric oxide on myocardial cell injury after hypoxia in rats
Objective To clarify the effect of NO on hypoxic myocardium in rats. Methods Myocardial cells from neonatal Wistar rats aged 1-2 days cultured for 4-6 days
then pretreated with GSNO (30
200
4 000 μmol/L) were exposed to normoxia (N) or hypoxia (H)(1%O
2
5%CO
2
and 94%N
2
) for 4 h. The NO content and LDH in the supernatant were detected. Results NO of 1.43
7.2 and 70 μmol/L were detected respectively in the supernatant of hypoxic myocardial cells treated by GSNO (30
200 and 4 000 μmol/L). Compared with normoxic group
LDH activity increased and myocardial cell activity decreased markedly in H+G30 and H+G4000 groups but les
s injury was observed in H+G200 group. Conclusion NO of certain concentration can markedly abate myocardial cell injury induced by hypoxia.
