摘要

目的最初报道的iASPP长度为351个氨基酸，但最近发现iASPP的全长应为828个氨基酸，为了探讨全长iASPP的功能，我们采取3分段扩增的策略克隆全长iASPP基因。方法从HL-60细胞中提取总RNA、DNA

RT-PCR分段扩增目的基因，克隆至pMD19-T simple载体，测序，并亚克隆至pCDNA3.1载体。结果重组真核表达质粒pcDNA3.1(+)-iASPP经测序，与GenBank上登陆的人iASPP cDNA（gi:60457962）序列完全一致。结论成功构建了iASPP真核表达载体。

Abstract

Objective iASPP was originally reported as long as 351 amino acids. Now

it is clear that the full-length of iASPP is as long as 828 amino acids. To investigate the function of full-length iASPP

we use three-step PCR technique to clone the full-length CDS of iASPP. Methods The total RNA was extracted from HL-60 and cDNA was amplified by RT-PCR. Then the amplified PCR product was digested and inserted into pMD19-T simple vector and subcloned into pCDNA3.1 vector. Results The gene sequence of iASPP was consistent with that reported (gi:60457962) in GenBank with cDNA sequencing. Conclusion The recombinant eukaryotic expression vector containing CDS region of iASPP has been constructed successfully.
